Tthe effect of recombinant interleukin-1 beta (rhIL-1 beta) and interleukin-8(rhIL-8) on bovine neutrophil degranulation was investigated by using myeloperoxidase (MPO), lactoferrin, and a 92-kDa gelatinase as the degranulating indicator for primary, secondary, and tertary granules, respectively. The release of MPO and lactoferrin was induced by rhIL-8 in a dose-dependent mannaer. Compared to the control, rhIL-1 beta only had a significant effect of the release of MP0 when the highest concentration was used (100 ng/ml). This effect was augmented when rhIL-1 beta solution was preincubated with MAC-T cells for 4 h. In contrast, the release of gelatinase was not altered by both cytokines. These results indicate that rhIL-8 has a dose effect on bovine neutrophil degranulation of primary and secondary granules; whereas rhIL-1 beta only has a significant effect at high concentrations. In addition, bovine mammary epithelial cells (MAC-T) might generate other degranulating factors in response to rhIL-1 beta.
