Species recognition of spider mites is difficult due to their small body sizes and unstable morphological characters in the immature stages. In this study, sequences of 28S rDNA and ITS2 of ~2900 and 600 bp in length, respectively, were used to identify the molecular variations of five species of spider mites in four genera: Oligonychus coffeae (Nietner), Panonychus citri
McGregor, Pan. ulmi (Koch), Petrobia harti (Ewing), and Tetranychus kanzawai Kishida. Specific primers and restricted digestion enzymes were designed from the 28S rDNA and ITS2 regions, and applied for multiplex PCR and PCR-RFLP to identify these spider mites. Results showed that the
application of the two molecular markers was useful for identifying these economically important pests.
