Solid state fermentation employing bamboo tray as vessel has been adopted as the traditional way of red mold rice production for hundreds years. However, this method implies disadvantages of long fermentation time, labor intensive, quality inconsistency, and large space needed. Recently, submerged fermentation is studied with its feasibility on mass production of red pigment in a shorter fermentation time, though the contents of secondary metabolites is comparatively less than those prepared by the traditional way.
In this investigation, Monascus purpureus BCRC 31499 was used to study the optimal submerged fermentation conditions for production of red pigment, as well as secondary metabolites such as monacolin k and citrinin, using Taguchi method. Orthogonal array and signal to noise (S/N) ratio were adopted for experimental design, data analysis, and determination of influential factors, respectively. A 4-factor and 3-level, L9(34) design consisting of substrates (indica rice, sorghum, and barley flake), inocula (2, 6, and 10%), agitation speed (350, 450, and 550 rpm), and cultivation time (36, 48, and 60 hours) was employed. All the trials were conducted in a 5 L jar fermentor. The optimal combination was found based on the Taguchi data analysis, which was barley flakes for substrate, 10% inoculums, 550 rpm for agitation speed, and 60 hours fermentation. Verification tests showed that submerged fermentation with this optimal combination also produced the highest red pigment content. This combination was therefore determined as the optimal condition for red pigment production using Monascus purpureus BCRC 31499.
