The study was to investigate the age-associated transitions in sheep immunity. Animals were equally divided into juvenile, adult and elderly groups, housed in same environment and fed with a uniform diet. Evaluations on the innate immunity and adaptive immunity were performed. For evaluating innate immunity, the phagocytosis activity, neutrophils migrations, and production of bactericidal reactive oxygen species (ROS) were conducted. The antibody titer, delayed type hypersensitivity (DTH) and lymphoblastogenesis were assessed for adaptive immunity. The results showed that the elderly group had the highest production of intracellular ROS in polymorphonuclear cells isolated from peripheral blood when the juvenile group only exerted minimal effects. Monitoring phagocytosis activities at 3 h and 24 h post fluorescent labeled yeast inoculation ex vivo, the younger group had better phagocytosis activity at 3 h than that of elderly group, while there was an inverse result at 24 h. The juvenile group had the lowest DTH response at 24 h and the skin swellness resolved in a faster manner monitoring over a 72 h time frame. Moreover, the elderly and adult groups had similar DTH response at 24 h. The antibody titer was higher in mature groups than that of juvenile group.
