Ganoderma lucidum has been one of the most highly valued medicinal fungi for China for several centuries. Polysaccharide extracts from Ganoderma lucidum have been reported to exhibit multiple biological functions. Here, we report the effects of Ganoderma lucidum polysaccharides (GLP) on the liver metabolism and the anti-inflammatory responses of broilers. Dietary supplementation with various concentrations of GLP promotes increased body weight and feed efficiency. To evaluate the effects of GLP on immune responses, two different inflammatory induction methods have been demonstrated: microorganismal infection and chemicals injection. We observed a moderate improvement in body weight with the inclusion of GLP in the diet before and after challenge with microorganisms. Birds injected with croton oil exhibited decreased body weight gain and increased the ratio of liver weight to body weight. The results of a microarray data analysis showed that the pathway of acute inflammatory responses was highly significant upregulated in birds that were injected with croton oil and fed the control diet. Half of the genes in the acute inflammatory response pathway were activated by croton oil injection. However, GLP treatment significantly increased the body weight gain after croton oil injection and moderated the increased gene expression. Taken together, the administration of GLP improved the growth performance and moderated the expression of acute inflammatory proteins. In this article, a novel relationship between the use of GLP as a feed additive and the anti-inflammatory properties has been described. These results may help to elucidate the immuno-regulatory effects of fungal polysaccharides and may provide different methods for evaluating alternatives to antibiotic feed additives.
