Beta-Glucans are broadly used as a source of dietary fibers, notably found in barley and oats, in a highly viscous soluble form, or as a health food product component as an extract from mushrooms, due to their immunomodulatory effects. In the present study, sBG were extracted from Auricularia polytricha (AP), Clitocybe maxima (CM), Rigidoporus ulmarius (RU), Pleurorus citrinopileatus Sing (PC), Pleurotus ostreatus (PO) and Pholioia nameko (PN) by using 95% ethanol. They were chemically characterized with molecular weight (M.W.) distributions as following: 40-60% of sBG with M.W. about 143 kd and less than 20% of molecules with M.W. >636 kd or <10 kd with an exception of sBG extracted from PN, which molecules over 90% were around 660 kd. All of the sBG effectively increased cell activities of murine splenocyte and NK cell as compared with untreated cells. In addition, sBG extracted from PO and PN showed the highest immuno-stimulating activities on enhancing the cytotoxicities of NK cells in vitro than other sBG or laminarin, a commercial sBG as the positive control in this study. These results imply that sBG extracted from AP, CM, RU, PC and PO by using 95% ethanol contains most beta-glucans with M.W. of 143 kDa while PN extraction contains most sBG with M.W. around 660 kDa. Moreover, sBG extracted from these fungi may act as an effective immuno-stimulating agent to splenocytes and NK cells.
