Cryopreservation has been proposed as one of the most promising long-term conservation approach for banana which is today most exclusively mass propagated via the in vitro techniques. Highly proliferating banana meristem clusters have previously been cryopreserved using a vitrification procedure but the post-thaw recovery of the stored material was highly genotype-dependent and was proven especially low for the cultivars of AA and AAA genomic constitutions. Since vitrification is a multi-steps procedure, its improvement will rely on the time-consuming optimization of the numerous treatment steps. The present study thus has the objective to analyze the performance of banana meristem clusters at each treatment step of an entire cryopreservation cycle using a combination of biochemical (e.g. ascorbate peroxidase, catalase, polyphenol oxidase, superoxide dismutase, peroxidase, malondialdehyde) and viability (weight variation, tissue regrowth) markers, in an attempt to determine the most stressful steps of the procedure. Results showed that there was generally a good correlation between the biochemical and viability markers and that the intensity of stress progressively increased as the explants went through the cryogenic cycle. Exposure of the explants to the highly concentrative vitrification solution has especially resulted in poor regrowth and poor enzymatic responses of the explants and thus was considered as the most critical stage in the cryogenic cycle. It is believed that the optimization of the abovementioned stage would lead to an improved post-storage recovery of the banana meristem clusters.
