In this study, detecting doped concentration of milk powder doped fresh milk employing side-polished plastic fiber sensor is presented. Different dimensions and roughness of sensing area of plastic fiber sensor are considered and manufactured by fiber side-polished system. The side- polished plastic optical fiber sensor is intrinsic fiber sensor and detects the variable concentrations of sample by the corresponding refractive index changes of sample. Therefore, the changes of optical power incident into one fiber endface of a fiber sensor and measured at the other fiber endface can be related to the concentration changes of sample. The limit of detection of the side-polished fiber sensor is tested by a standard sucrose solution with the refractive indices in the range of 1.333 to 1.403. The best limit of detection is 1.53×10-4 RIU for refractive indices in the range of 1.333 to 1.403 and the sensing area dimensions are depth of 0.4 mm and length of 4.7 mm. The side-polished plastic fiber sensor used to detect fresh milk doped with different proportional milk powder, and four kinds of fresh milk doped with 0, 10, 20, and 50% milk powder are detected. For the detection sensitivity, all the samples are diluted one thousand times for the detection of fresh milk doped powder milk. The detected optical power through fiber sensor is positively correlated with the doped concentration of milk powder. In addition, three different high-power light emitting diodes with blue (λ = 460 nm), green (λ = 528 nm), and red (λ = 624 nm) color is also used in measurement. The detecting results done by blue light emitting diodes show the best slope and limit of detection are 0.1352 μw / % and 1.1646 % with linearity R = 0.9946 for milk powder doped concentration in the range of 0 to 50 %.
