In this study, detecting fresh milk doped milk powder employing side-polished plastic fiber sensor is presented. The side-polished plastic optical fiber sensor is intrinsic fiber sensor and detects the variable concentrations of sample by the corresponding refractive index changes of sample. Therefore, optical power incident into one fiber end face of a fiber sensor and the measured optical power changes at the other fiber end face can be related to the concentration changes of sample. The sensor resolution of the side-polished fiber sensor is tested by a standard sucrose solution with the refractive indices in the range of 1.333 to 1.403. The best sensor resolution is 1.53×10-4 RIU. The side-polished plastic fiber sensor used three different high-power light emitting diodes with blue (λ = 460nm), green (λ = 528nm), and red (λ = 624nm) color in measurement. The fresh milk doped with milk powder of four concentrations set to 0, 10, 20, and 50% as well as whey protein samples produced by a centrifuge are detected. The detecting results show that the best sensor resolution of 1.1646% is done by blue light source and the doped concentration of milk powder in the range of 0 to 50 %.
