Apoptosis plays an important role in infection, cancer, embryo development and tissue growth balance. In intracellular mechanism of infection avoiding, apoptosis could be induced to prevent intracellular proliferation of virus or bacteria. Apoptosis included caspase-dependent and caspase-independent pathways. Due to the previous studies revealed that the caspase-dependent pathway is not the only one apoptotic pathway in Salmonella-infected cells, the other apoptotic pathway in Salmonella-induced apoptosis needs to be manifested. The TUNEL and Annexin V assay on mice tissues to evaluate apoptosis, and immunohistochemistry and immunofluorescence on mice tissues to evaluate the effect of AIF, Endo G and caspases. After the cells were treated with Salmonella, the Salmonella-infected cells were treated with caspase inhibitor for the studies of TEM and immunofluorescence to analyze the intracellular distribution of Salmonella. We realized the intracellular effects of AIF and Endo G and the relationships between them by Western Blot analysis. Thus, we clarify Salmonella induced apoptosis via caspase-independent pathway. The changes and interaction of apoptotic proteins, AIF and Endo G. in Salmonella- induced apoptosis.
